Chemical Genomic Study of Endocrine Disrupting Chemicals in Metabolic Pathways
نویسندگان
چکیده
Endocrine disrupting activities in industrial chemicals have been a serious problem, because they are distributed broadly, they are active at very low concentrations, and some of them are difficult to detoxify. Moreover, only a small number of active chemicals are known, while most chemicals have not been examined yet. To reduce the number of examinations required for unknown chemicals, some computational prediction methods have been proposed. In this study, we propose a chemical genomic approach to understand the relationship between activities and chemical structures. Our approach revealed the existence of synthetic and degradation chains in metabolic pathway maps. At the end of the degradation chains, we found certain oxidization enzymes that appeared frequently. We also confirmed our prediction by performing a bioassay experiment. The results would help us to find potential detoxifying microorganisms. In addition, our method can be applied to other types of toxic chemicals, such as cancer-causing agents.

Endocrine disrupting chemicals targeting estrogen receptor signaling: identification and mechanisms of action.
Many endocrine disrupting chemicals (EDCs) adversely impact estrogen signaling by interacting with two estrogen receptors (ERs): ERα and ERβ. Though the receptors have similar ligand binding and DNA binding domains, ERα and ERβ have some unique properties in terms of ligand selectivity and target gene regulation. EDCs that target ER signaling can modify genomic and nongenomic ER activity throug...

Concerns about children's weight have steadily risen alongside the manufacture and use of myriad chemicals in the US. One class of chemicals, known as metabolic disruptors, interfere with human endocrine and metabolic functioning and are of specific concern to children's health and development. This article examines the effect of residential concentrations of metabolic disrupting chemicals on c...
متن کاملIs exposure to environmental or industrial endocrine disrupting estrogen-like chemicals able to cause genomic instability?
Human and wild life populations are continually exposed to a wide variety of environmental estrogen-like chemicals. Most research to date on environmental endocrine disrupting estrogen-like chemicals has focussed on screening of estrogenic activity of environmental or industrial chemicals, their bioaccumulative properties and toxicokinetics, and developing the structure-activity relationship be...
